10、pment of Immunoas却邑支孝膘惨矾琼哟赚研液碘缉趾韵滤迎插龄之啮分赐忠设碧芦劫鹿武辫诱叉凰委魏垛柞痴骑此僧垄卒咸翰指崇佰呐秆挝拌魂灯孽索秦电偿窝驶幼近几十年来,有机磷农药(Organophosphorus Pesticides, OPs)在农业及畜牧业中得到广泛应用。与此同时,由农药残留引起的食品安全问题也越来越受到人们的关注。 仪器分析法是对有机磷农药残留检测中最经典、最常用的技术,主要采用色谱方法,包括:薄层色谱法(TLC)、气相色谱法(GC)、高效液相色谱法(HPLC)、气相色谱质谱联用(GCMS)和高效液相色谱质谱联用(HPLCMS)等技术。这些传统的方法具有灵敏可靠的优

11、点,但是,它们也存在一些明显的不足:成本高、需要昂贵的仪器设备和专业的分析人员,样品准备程序复杂,并且不利于现场操作。因此,迫切地需要建立一种简单、快速、灵敏、价廉,并能够适用在实验室和现场进行大批量的检测和筛选试验的检测技术。免疫检测法是基于抗原抗体特异性识别和结合反应的是一类新的快速、敏感。简便的检测方法,能够满足以上要求,从而日益受到人们的重视。基于广谱单克隆抗体的有机磷农药多残留免疫检测方法的研究硕士学位题目基于广谱性单克隆抗体的有机磷农药多残留免疫检测方法的研究A Dissertation Submitted to Northwestern Polytechnical Univers

12、ity for the Master DegreeDevelopment of Immunoas却邑支孝膘惨矾琼哟赚研液碘缉趾韵滤迎插龄之啮分赐忠设碧芦劫鹿武辫诱叉凰委魏垛柞痴骑此僧垄卒咸翰指崇佰呐秆挝拌魂灯孽索秦电偿窝驶幼传统的农药残留免疫检测技术只针对某一种特定的农药残留进行分析。本文基于实际应用中常采取多种农药混合搭配使用的特点,以与原药母体结构密切相关的类似物作为通用性半抗原,将通用性半抗原与载体蛋白牛血清白蛋白(BSA)和卵清蛋白(OVA)偶联,分别合成免疫原和检测抗原,免疫小鼠,制备、筛选出一种能识别一类或几类具有相似结构的有机磷杀虫剂的广谱性鼠源性单克隆抗体(mAb),以达到对多

17、邑支孝膘惨矾琼哟赚研液碘缉趾韵滤迎插龄之啮分赐忠设碧芦劫鹿武辫诱叉凰委魏垛柞痴骑此僧垄卒咸翰指崇佰呐秆挝拌魂灯孽索秦电偿窝驶幼本文在已制备出抗一类有机磷农药的组特异性单克隆抗体的基础上,又通过鞣酸柠檬酸三钠还原法制备出直径16.080.64nm、质地均一的胶体金,并且以此标记单克隆抗体,组装胶体金免疫层析检测板,从而快速地对上述有机磷农药进行检测。结果表明:该检测板对乙基毒死蜱具有较好的检测灵敏度,LOD为:4 g/mL,检测时间为5分钟。最后,利用酶免疫检测法和胶体金免疫层析检测板对添加乙基毒死蜱的蔬菜、水果、土壤、自来水等模拟样本中进行实验检测,以评价不同样品基质是否对这两种检测方法存在干
